Contents
Per 5 mL Co-amoxiclav (amoxicillin & clavulanic acid): Amoxicillin trihydrate 400 mg, clavulanic acid 57 mg
Indications/Uses
Upper resp tract infections (including ENT), in particular sinusitis, otitis media, recurrent tonsillitis; lower resp tract infection, in particular acute exacerbation of chronic bronchitis & bronchopneumonia; genital & UTI; skin & soft tissues infections.
Dosage/Direction for Use
Adult 1,760 mg/day (or 11 mL bid). Childn 2 mth-12 yr Mild to moderate infections (skin & soft tissue infections) 25/3.6 mg/kg/day. Severe infections (otitis media, sinusitis & lower resp tract infections) 45/6.4 mg/kg/day.
Administration
Should be taken with food: Best taken at the start of meals to reduce GI discomfort.
Contraindications
Hypersensitivity to penicillins. History of severe immediate hypersensitivity reaction (eg, anaphylaxis) to another β-lactam agent (eg, cephalosporin, carbapenem or monobactam). History of jaundice/hepatic impairment.
Special Precautions
Previous hypersensitivity to penicillins, cephalosporins or other β-lactam agents. Do not use on penicillin-resistant Strep pneumoniae. Convulsions may occur in patients w/ impaired renal function or in those receiving high dose. Avoid if infectious mononucleosis is suspected. Concomitant use w/ allopurinol. Prolonged use may result in overgrowth of non-susceptible organisms. Discontinue if feverish generalised erythema associated w/ pustula occurs. Antibiotic-associated colitis. Periodic assessment of organ system functions including renal, hepatic & hematopoietic function is advisable during prolonged therapy. Maintain adequate fluid intake & urinary output in order to reduce possibility of amoxicillin crystalluria. False +ve w/ non-enzymatic methods & Coombs' test. Cross-reactions w/ non-Aspergillus polysaccharides & polyfuranose. Hepatic impairment. Avoid use during pregnancy. Lactation.
Adverse Reactions
Mucocutaneous candidosis; diarrhea, nausea, vomiting.
Drug Interactions
Increased likelihood of allergic skin reactions w/ allopurinol. Increased INR in patients maintained on acenocoumarol or warfarin. May reduce excretion of methotrexate. Decreased renal tubular secretion of amoxicillin w/ probenecid. Reduction in pre-dose conc of the active metabolite mycophenolic acid.
